It is of great significance to systematically trace the transmission and acquisition
process in children for Streptococcus mutans which has been attributed as the chief
causative organism for dental caries. This study was performed in order to examine the
difference or similarity of genotypes of oral S. mutans in 2¡4 year old infants and their
mothers by using the DNA finger-printing with gtfB gene probe. From stimulated whole
saliva, S. mutans was isolated and selectively cultured, from which chromosomal DNA
was isolated. Isolates of chromosomal DNA was treated with restriction enzyme, EcoRI
and BamHI, hybridized with labelled DNA probe, and processed to obtain DNA
fingerprinting patterns. The site where chromosomal DNA and labelled DNA probe
bound represented a band, the location of which was different individual to individual or
pair to pair of mother and her child.
The results obtained were as follows.
1. Streptococcus mutans was not isolated in some children, and the tendency was
stronger in younger children (3/12).
2. Among 9 mother-child pairs, 6 pairs showed the identical DNA fingerprinting
patterns. The identity was not related to the chronological age of the children.
3. It was confirmed that DNA fingerprinting with gtfB gene probe is valuable for the
epidemiological study on the nature of transmission of the cariogenic microorganism, S.
mutans.
